Ion-Channel Gating: Twist to open
نویسنده
چکیده
Three-dimensional images of the open state of the nicotinic acetylcholine receptor have been obtained at 9 A resolution. Comparison with the closed state reveals the structural basis of channel gating.

Inherent Dynamics of the Acid-Sensing Ion Channel 1 Correlates with the Gating Mechanism
The acid-sensing ion channel 1 (ASIC1) is a key receptor for extracellular protons. Although numerous structural and functional studies have been performed on this channel, the structural dynamics underlying the gating mechanism remains unknown. We used normal mode analysis, mutagenesis, and electrophysiological methods to explore the relationship between the inherent dynamics of ASIC1 and its ...
متن کاملInhibition mechanism of the acetylcholine receptor by alpha-neurotoxins as revealed by normal-mode dynamics.
The nicotinic acetylcholine receptor (AChR) is the prototype of ligand-gated ion channels. Here, we calculate the dynamics of the muscle AChR using normal modes. The calculations reveal a twist-like gating motion responsible for channel opening. The ion channel diameter is shown to increase with this twist motion. Strikingly, the twist motion and the increase in channel diameter are not observe...
متن کاملGating Behavior of Endoplasmic Reticulum Potassium Channels of Rat Hepatocytes in Diabetes
Background: Defects in endoplasmic reticulum homeostasis are common occurrences in different diseases, such as diabetes, in which the function of endoplasmic reticulum is disrupted. It is now well established that ion channels of endoplasmic reticulum membrane have a critical role in endoplasmic reticulum luminal homeostasis. Our previous studies showed the presence of an ATP-sensitive cationic...
متن کاملSolution-Based Single-Molecule FRET Studies of K(+) Channel Gating in a Lipid Bilayer.
Ion channels are dynamic multimeric proteins that often undergo multiple unsynchronized structural movements as they switch between their open and closed states. Such structural changes are difficult to measure within the context of a native lipid bilayer and have often been monitored via macroscopic changes in Förster resonance energy transfer (FRET) between probes attached to different parts ...
